Often, but not always, the symptoms you describe are caused by a corrupt print job stuck in the queue or a damaged printer driver. However before you clean things up, on general principles, that you can download, install, update and run full scans with each of these two free programs:
AntiMailware MalwareBytes
SUPERAntiSpyware
Do not operate the two scans simultaneously. Each will take a long time, so start it and then go do something else for a while.
Cleaning of printers
NOTE: If after completing step has the spooler print don't rest not always running after you launched the command "net start spooler", follow these steps
Open a command prompt window (start > run > cmd > OK)
Type the following line in the black command prompt window
dependent on the spooler of sc config = RPCSS
net start spooler
If the spooler still will not start, you will not be able to follow all the steps in "First Article". Instead, go to the other link and download and get and use the utility 'cleanspl' such as described here.

Open Services by typing "Services.msc" in the Run command window and clickin on 'Ok '.
Double click on "Printer Spooler" Services listed there.
Click on stop, click OK and go to '% WINDIR%\system32\spool\printers' and all delete this folder.
Now again go to Services.msc and start the Printer Spooler service and change the startup as "Automatic" Type
Click Ok and restart the computer and check.

I have a Dell Dimension desktop E510 and a Brother MFC-465CN printer. Unable to add my printer as the print spooler service is not running. FYI: I had a virus a week ago and added Malwarebytes' Anti-Malware to correct the problem in case it is of any relevance. Try to reset the spooler Services service, click Start-> Run Type & -> services.msc-> ok.
Locate the printer spooler on the right side-> right click and select Restart-> now Double click on Print spooler-> select automatic for startup type.
If give you the error, try to replace spoolsv.exe is located in C:\windows\system32 in C:\WINDOWS\ServicePackFiles\i386.

I have a PC running on Windows Vista professional that I had for about a year with two printers that have also been all this time, a HP LaserJet 1000 and an Epson SX105.
This morning, amid a series of PDFs (separate files) printing on LaserJet printers, I suddenly had the following pop-up message:
"Spooler SubSystem App stopped working and was closed".
On the Control Panel, I found that my two printers have apparently disappeared. My first reaction was just close and restart the computer, often kind of seeds.
To restart, I tried to print again, but I was told that I had to install a printer. Trying to install a printer, I learned that I could not because of a problem with the spooler. I looked online to find a solution and one that suits (without touching the registry) seemed to be to check the operation of via Control Panel's print spooler, and then restart. Oh dear. What a mistake! I did this and found that the printers have reappeared, but then about 30 seconds later, I got the following pop-up message:
"Windows must now restart because the print spooler service terminated unexpectedly.
Each time it restarts the message comes back after less than a minute and restarts on another 30 seconds later, so I have a chance to try a system restore (I installed the last service pack Vista a couple of days), or something else... it's just in a loop of perpetual reboot, I can't stop restarting Windows.
How the hell do I? (I send this message using my Vista laptop and pray the same problem occur here, too, so I don't have to print anything).
